Adult Dosing

  • 2-4g PO daily in 1-4 doses

Pediatric Dosing

  • >2mo: 75-150 mg/kg PO daily in 1-4 doses

Contraindications

  • Allergy to class/drug
  • Infants <2mo (except to treat congenital toxoplasmosis)
  • Pregnant women at term
  • Breastfeeding

Adverse Reactions

Serious

  • SJS/TEN
  • Agranulocytosis, hemolytic anemia, leukopenia, thrombocytopenia, aplastic anemia,
  • Crystalluria

Common

  • Rash
  • Nausea/vomiting, diarrhea, abdominal pain

Pharmacology

  • Half-life: 7-16.8h
  • Metabolism: Hepatic
  • Excretion: Mostly renal

Mechanism of Action

  • Inhibits bacterial synthesis of folic acid
